The effect of thiamine deficiency on the glucose oxidative pathway of rat erythrocytes.

چکیده
Thiamine pyrophosphate has been demonstrated recently to be an essential cofactor for the transketolation of pentose to heptulose in the hexose monophosphate shunt in addition to its well known function in pyruvate utilization by animal tissues (l-4). The observation that the glucose oxidative pathway of mammalian erythrocytes may be studied in vitro in the absence of interfering oxidative phenomena by the addition of methylene blue (5) made it possible to determine the effect, if any, on the transketolase reaction by acute thiamine deficiency, in this tissue. The data to be presented demonstrate that rat erythrocyte transketolase activity is reduced by the removal of thiamine from the diet. This lends further support to the argument that methylene blue activates the glucose oxidative pathway in erythrocytes, and, in addition, furnishes an assay for evaluation of thiamine nutrition of animals.

Protective Effect of Quercetin on Oxidative Stress in Glucose-6-Phosphate Dehydrogenase-Deficient Erythrocytes in Vitro
Glucose-6-phosphate dehydrogenase (G6PD) deficient subjects are vulnerable to oxidative stress. Quercetin, a flavonoids, has been employed as a potent oxygen-free radical scavenger in order to assess the protective effects of quercetin against H2O2-induced oxidative damage in G6PD-deficient and normal human erythrocytes. Erythrocytes of G6PD-deficient (n = 10) and normal (n = 10) subjects were ...

متن کاملEffect of thiamine deficiency, pyrithiamine and oxythiamine on pyruvate metabolism in rat liver and brain in vivo.
Rats were fed either a thiamine-deficient diet of diets containing pyrithiamine or oxythiamine. When symptoms of thiamine deficiency appeared, the animals were injected intraperitoneally with [2-14C] pyruvate six to twelve minutes prior to sacrifice. Free glutamic and aspartic acids were isolated from liver and brain and degraded. The results indicate that, in thiamine-deficient or oxythiamine-...
متن کاملAntioxidant Enzymes and Oxidative Stress in the Erythrocytes of Iron Deficiency Anemic Patients Supplemented with Vitamins
Background: Iron deficiency anemia is one of the major causes of morbidity and mortality worldwide. Evidences from epidemiological and clinical studies suggest a possible correlation between antioxidant levels and the anemic disease risk. The present work is to investigate antioxidant levels and lipid peroxidation in anemic patients. Methods: A number of 30 patients (15 males and 15 females) we...
متن کاملThe effects of Ziziphus jujuba fruit extract on catalase activity and lipid peroxidation in the heart and erythrocytes of rats following chronic ethanol consumption
Chronic ethanol consumption leads to oxidative stress in the heart and erythrocytes of rats. As Ziziphus jujuba fruit has been shown to have potent antioxidants, such as flavonoids, we conducted this study to evaluate the effects of aqueous fruit extracts from Z. jujuba on rat hearts and erythrocytes following chronic ethanol consumption. Twenty-eight male Wistar rats were randomly divided into...
